Establishment and Evaluation of an Animal Model of Dilated Cardiomyopathy

摘要: 目的 探索建立扩张型心肌病(DCM)动物实验模型的方法并对其进行评估,为 DCM 的临床诊断治疗提供思 路与方法。 方法 实验选择成年比格犬,分为两组,实验组注射阿霉素,对照组注射等量 0. 9% 氯化钠溶液。 经麻 醉诱导进行冠状动脉造影后,经左侧冠状动脉主干注射阿霉素(多柔比星) ,每周注射一次,连续注射 5 周。 建立实 验组 6 只,对照组 4 只,末次注射 4 周后对各组动物进行心脏超声检测、多导生理记录仪及心导管技术检测、血液学 检测及心室重构评价检测,以评估 DCM 建模的可行性及有效性。 结果 与对照组相比,DCM 组实验动物心脏出现 明显扩大,且心室收缩力出现明显降低;DCM 组实验动物与对照组相比,各项血液学指标( 除白细胞计数外) 均出 现明显差异;与对照组相比,DCM 组实验动物出现明显地全心扩大,Collagen I / III 的含量明显升高。 结论 采用冠 脉多次注射阿霉素(多柔比星)的方法建造 DCM 动物模型,具有可行性及有效性,其周期较短、简单易行,并且更接近于人类 DCM 病理生理过程。

Abstract: Objective To explore the method of establishing and evaluating the animal experimental model of dilated cardiomyopathy (DCM) , and to provide ideas and method for the clinical diagnosis and treatment of DCM. Method Adult beagle dogs were divided into two groups, the experimental group was injected with adriamycin and the control group was injected with 0. 9% sodium chloride solution. Adult beagles were given doxorubicin ( doxorubicin) through the left main coronary artery once a week for 5 weeks after coronary angiography induced by anesthesia. The DCM group ( n = 6) was established, and the sham group ( n = 4) was established. Four weeks after the last injection, the animals in each group were examined by cardiac ultrasound, multichannel physiological recorder and cardiac catheterization technology, hematology and ventricular remodeling evaluation to evaluate the feasibility and effectiveness of DCM modeling. Result Compared with the control group, the heart of the experimental animals in the DCM group was significantly enlarged, and the ventricular contractility was significantly reduced. Compared with the control group, the experimental animals in DCM group showed significant differences in all hematological indexes ( except white blood cell count) . Compared with the control group, and the content of Collagen I / III in DCM group was significantly increased. Conclusion Multiple injection of doxorubicin into the coronary artery is feasible and effective to construct the animal model of DCM. The cycle is short, simple and feasible, and it is more similar to the pathophysiology of human DCM.
